Abstract Acrobustitis is the inflammation of the distal prepuce, which can lead to a narrowing of the preputial ostium due to stenosis or growth of fibrous tissue after an inflammatory reaction. This condition usually occurs in cattle with long prepuce, such as Zebu or Zebu’s crossbreeds, leading the animal to Impotentia Coeundi, this condition is characterized by the bull's disability to copulate, that leads to lower herd fertility and consequent financial losses. Normally, corrective surgeries are performed on-farm and the animal is placed in a lateral recumbency. However, in some situations the animal is restrained with ropes and remains on the grass, dirt or even on uneven floors, which can cause neuropathies, bloat or hypoxia. Due to a series of complications that can occur in the postoperative period of surgery in the lateral recumbency, this article aims to describe the surgical technique for correcting acrobustitis with the animal in a standing position. Ten corrective surgeries for acrobustitis were performed in adult bulls between 4 and 8 years of age and predominantly of zebu or crossbreeds, with a total recovery of the animals for full reproductive activity.

Abstract Sperm cryopreservation has become an indispensable tool in reproductive biology. However, frozen/thawed semen has a short lifespan due to loss of sperm cell integrity. To better understand which sperm cell structures are compromised by the cryopreservation process and apoptosis markers, the sperm of five healthy mature dogs was analyzed in this study. Analysis was performed after collection, cooling, and thawing via computer assisted sperm analyzer (CASA) and evaluation of membrane fluidity and permeability, phosphatidylserine translocation (Annexin V), membrane integrity, mitochondrial membrane potential, membrane lipid peroxidation (LPO) and activity of the apoptotic markers caspases 3 and 7 by flow cytometry. Cryopreservation decreased total and progressive motility and the percentage of rapid sperm (P < 0.01). Damage to sperm cells was confirmed by Annexin V (P <0.01), indicating that capacitation-like changes were induced by the cryopreservation procedures. An increase in sperm membrane fluidity was also noted in frozen/thawed samples (P < 0.01). Plasma and acrosomal cell membranes were affected (P < 0.01), with decreases in the subpopulation displaying high membrane potential (P < 0.01). Membrane LPO was increased in thawed sperm compared to cooled sperm (P < 0.05) but was not different from that in fresh sperm. No differences were observed in caspase 3 and 7 activity after cooling, freezing, or thawing. In conclusion, total and progressive motility, plasma membrane integrity and mitochondrial membrane potential suffered from the deleterious effects caused by cryopreservation, unlike the activity of caspases that remained stable during the freezing process.

Descreve-se um caso de infertilidade de um jumento SRD confirmada por meio de microscopia eletrônica de transmissão (MET). O espermiograma, avaliado sob microscopia ótica, revelou baixa motilidade e alta concentração de anormalidades espermáticas do tipo gota citoplasmática proximal. O material foi avaliado por MET, observando-se um acúmulo desordenado de microtúbulos causando protusões irregulares na região do colo espermático. O último teste realizado correspondeu ao de fertilidade in vivo, utilizando-se quatro éguas portadoras de bom histórico reprodutivo, nas quais não foi possível confirmar nenhuma prenhez. Frente aos resultados obtidos, associados aos achados da MET, estabeleceu-se o diagnóstico de infertilidade associada a defeito microtubular dos espermatozóides.

A donkey infertility was described by transmission electron microscopy (MET). The spermiogram evaluated by light microscopy showed low sperm motility and high concentration of abnormal sperm with proximal cytoplasmic droplets. The material was evaluated by MET, where it was observed disarrangement of microtubules, causing irregular protrusions in the spermatic neck. The last test done was the in vivo fertility, using four mares, with a reproductive healthy historic, where no pregnancy occurred. Facing the results that we had in vivo, associated with the MET findings, we diagnosed infertility associated with microtubular defect of the spermatozoa.

O objetivo deste trabalho foi avaliar, in vitro, o efeito da adição de 1 mg pentoxifilina por mL de amostras seminais diluídas em meio Kenney, e resfriadas a 5º C, de garanhões normospérmicos, sobre os seguintes parâmetros: taxas de motilidade total e motilidade progressiva, velocidade espermática ao longo da trajetória real, índice retilíneo, taxa de linearidade, velocidade progressiva, taxa de viabilidade e taxa de integridade da membrana plasmática. Foram avaliados 20 ejaculados de quatro garanhões, com o auxílio do Hamilton Thorn Research (Animal Version 12.0 L, EUA), em câmara de Makler. A taxa de viabilidade foi realizada por coloração de eosina e microscopia de contraste e a taxa de integridade de membrana, por coloração com fluorocromos, iodeto de propídio e diacetato de carboxifluoresceina e microscopia de fluorescência. As análises foram realizadas aos 30, 60, e 120 minutos de incubação a 37º C, após 12, 24 e 48 horas de resfriamento a 5ºC. A pentoxifilina incrementou significativamente, em relação ao grupo controle, os parâmetros espermáticos relacionados à motilidade dos espermatozóides e de integridade da membrana plasmática, durante todo o período de incubação, da seguinte forma: taxa de motilidade total em 8,2%; motilidade progressiva em 4,7%; velocidade real em 23,1 mm/s; taxa de espermatozóides com velocidade rápida em 7,4%; taxa de viabilidade em 4,7%; e integridade da membrana plasmática em 3,5 % do tratado. Quanto aos parâmetros índice de retilinidade e linearidade, não foi observada diferença significativa entre os grupos. Os resultados obtidos nas condições deste experimento indicam que a adição de pentoxifilina ao ejaculado resfriado de garanhões, até 48 horas após o resfriamento, pode ser benéfica à qualidade dos espermatozóides para uso na inseminação artificial.

The objective of this work was to evaluate the in vitro effect of the addition of 1 mg pentoxifylline per ml of seminal samples from normospermic stallions, which were first diluted in Kenney extender and cooled at 5ºC. The evaluation was carried out using the following parameters: total and progressive motility rate, track speed, straightness and linearity rate, progressive velocity, viability rate and plasmatic membrane integrity rate. For this experiment, 20 ejaculates from four stallions were examined using Hamilton Thorn Research (Animal Version 12.0 L, EUA) with a Makler camera. The viability rates were obtained using eosin dyes under a light microscope and membrane integrity was obtained by fluorochrom, propidium iodine and carboxyfluorescein diacetat dyes under a fluorescence microscope. The semen examination occurred at 30, 60 and 120 minutes of incubation at 37º C, after 12, 24 and 48 hours of cooling at 5ºC. The pentoxifylline significantly increased the sperm parameter rates of motility and plasmatic membrane integrity for all incubation times, when compared with the control group. The rates were: +8.2% total motility, +4.7% progressive motility, +23.1 mm/s track speed, +7.4% progressive velocity, +4.7% viability and +3.5% plasma membrane integrity. The straightness and linearity rates were not significantly different between the control and treated groups. The experimental data confirm that pentoxifylline addition to stallion-cooled sperm, up to 48 hours after cooling, could preserve sperm quality for artificial insemination.

O presente estudo objetivou diagnosticar as causas da morte embrionária precoce em 128 éguas. Amostras de soro foram coletadas nos dias 4, 7, 10, 13, 16, 19, 21 e 30 após ovulação, para dosagens hormonais e mensurações do corpo lúteo. O diâmetro e as características da vesícula embrionária foram avaliados, através da ultra-sonografia, a partir do 12º dia pós-ovulação. Das 128 éguas estudadas, 17 (13.28%) apresentaram morte embrionária. O diâmetro do corpo lúteo, bem como as concentrações plasmáticas de progesterona, foi semelhante nos grupos que apresentaram morte embrionária e nos que mantiveram a gestação. Os níveis de estrogeno plasmático foram mais elevados no grupo das fêmeas que mantiveram a gestação. Os exames citológicos, microbiológicos e histopatológicos revelaram que a maioria das éguas com diagnóstico de morte embrionária eram portadoras de endometrites.

The present study aimed to diagnose early embryonic death in 128 mares. Blood samples were collected at the 4th, 7th, 10th, 13th, 16th, 19th, 21st and 30th day after ovulation and the diameter of the corpus luteum was measured. The diameter and characteristics of the embryonic vesicle were evaluated by ultrasonography. Microbiology, cytology and histopathology were carried out in mares with embryonic death. Plasma oestrogen and progesterone concentration were measured by radioimmunoassay. From 128 mares, 17 (13.28%) showed early embryonic death. The embryonic losses took place at 19 days of pregnancy in 47.05% and at 21 days of pregnancy in 29.4% of the mares. The diameter of the corpus luteum in the mares that maintained pregnancy was similar to those with embryonic loss. Otherwise, the diameter of the embryonic vesicles was bigger at the 16th, 19th, 21st and 30th day of pregnancy when compared to the mares with embryonic loss. The mean plasma progesterone concentration was similar in both groups and the median plasma oestrogen concentration was higher in the pregnant mares. The cytological, microbiological and histopathological exams revealed that most of the mares had endometritis. Ultrasonography provided an early diagnosis of pregnancy (from the 12th day post-ovulation) and important information about the development of embryo and embryonic death. Endometritis was considered the main cause of embryonic losses in this study.
